Overview

ARC-T cells + SPRX002 (also known as ACLX-002) is a two-component, universal CD123-targeted cell therapy for the treatment of relapsed or refractory acute myeloid leukemia (AML) and high-risk myelodysplastic syndrome (MDS). ARC-T cells are autologous T cells that are genetically engineered to express an antigen-receptor complex (ARC) that can bind to a tag on SPRX002. SPRX002 is a soluble, monovalent protein adapter that binds CD123 on the surface of leukemia cells and acts as a bridge, enabling the targeted activation and cytotoxic function of ARC-T cells upon engagement. The system is modular: ARC-T cells are inactive until SPRX002 is administered and bridges them to CD123-expressing targets. This allows dosing and activity control, potentially improving safety and efficacy versus conventional CAR-T.[1][2][3][4][7][8]
