Drug intelligence / Profile preview
**argatroban + clopidogrel + aspirin** is a combination drug therapy consisting of three agents: - **Argatroban** is a synthetic, direct thrombin inhibitor (DTI) that inhibits both free and clot-associated thrombin, reducing fibrin formation, activation of coagulation factors V, VIII, XIII, protein C, and platelet aggregation. It is primarily used in patients at risk for heparin-induced thrombocytopenia and for anticoagulation in acute thrombosis[5][1][3]. - **Clopidogrel** is an oral thienopyridine class antiplatelet agent that irreversibly inhibits the P2Y12 ADP receptor on platelets, thereby preventing platelet activation and aggregation and reducing risk of arterial clot formation[4][2]. - **Aspirin** is a nonsteroidal anti-inflammatory drug (NSAID) with antiplatelet properties, irreversibly inhibiting cyclooxygenase (COX-1), thus preventing thromboxane A2 production, which suppresses platelet activation and vasoconstriction[2]. This combination targets both **thrombin activity and platelet activation/aggregation**, aiming for synergistic antithrombotic effects. It is primarily studied in acute ischemic stroke settings, such as single subcortical infarction (SSI), intracranial atherosclerosis (ICAS), and posterior circulation ischemic stroke (PCIS), with evidence suggesting benefit for neurological recovery and prevention of early neurological deterioration (END) without a significant rise in intracerebral hemorrhage risk when used short-term[1][3].
