Drug intelligence / Profile preview
ARIKAYCE (amikacin liposome inhalation suspension) is a liposomal formulation of the aminoglycoside antibiotic amikacin developed by Insmed Incorporated. It is specifically indicated for the treatment of Mycobacterium avium complex (MAC) lung disease in adult patients who have not achieved negative sputum cultures after at least six consecutive months of a multidrug background regimen. The drug utilizes proprietary PULMOVANCE liposomal technology to encapsulate amikacin in small liposomes (approximately 300 nm), which are delivered directly to the lungs via the Lamira Nebulizer System. This targeted delivery allows the drug to be taken up by alveolar macrophages, the primary site of MAC infection, while minimizing systemic exposure and the severe toxicities typically associated with intravenous amikacin, such as ototoxicity and nephrotoxicity. Amikacin works by binding to the bacterial 30S ribosomal subunit, disrupting protein synthesis and leading to bacterial cell death.
