Drug intelligence / Profile preview
Arikayce (amikacin liposome inhalation suspension) is an inhaled antibacterial agent developed by Insmed Incorporated, specifically designed to treat chronic lung infections. It utilizes a liposomal lung delivery technology to encapsulate the aminoglycoside amikacin, allowing for deep lung penetration and uptake by alveolar macrophages. This formulation is particularly effective against *Pseudomonas aeruginosa* and *Mycobacterium avium* complex (MAC) because the liposomes can penetrate bacterial biofilms that typically protect these pathogens from standard antibiotics. The mechanism of action involves amikacin binding to the bacterial 30S ribosomal subunit, which disrupts protein synthesis and leads to bacterial cell death. While it is FDA-approved for refractory MAC lung disease, it has been extensively studied in Phase 2 and Phase 3 trials for the treatment of chronic *Pseudomonas aeruginosa* infections in patients with bronchiectasis and cystic fibrosis.
