Aripiprazole + valproate is a fixed-dose combination (FDC) tablet formulation comprising the atypical antipsychotic aripiprazole and the mood stabilizer divalproex sodium (valproate). Aripiprazole acts as a partial agonist at dopamine D2 and serotonin 5-HT1A receptors and an antagonist at 5-HT2A receptors, providing stabilizing effects on neurotransmission. Valproate's mechanism involves increasing gamma-aminobutyric acid (GABA) levels in the brain and modulating voltage-gated sodium channels to stabilize neuronal activity. This combination is primarily indicated for the treatment of manic or mixed episodes associated with bipolar I disorder. The development of the FDC aims to improve patient compliance and simplify dosing regimens compared to the co-administration of individual tablets.
