ARO-ALK7 + tirzepatide is an investigational combination therapy under clinical evaluation for the treatment of obesity, including in populations with and without type 2 diabetes mellitus. ARO-ALK7 is an RNA interference (RNAi) therapeutic designed to silence the ACVR1C gene, thereby reducing expression of Activin receptor-like kinase 7 (ALK7) in adipose tissue to promote lipid mobilization, increase catecholamine sensitivity, and reduce fat mass while preserving lean mass. Tirzepatide is a dual agonist of the glucagon-like peptide 1 (GLP-1) receptor and glucose-dependent insulinotropic polypeptide (GIP) receptor, approved as a monotherapy for type 2 diabetes and obesity, that improves glycemic control and reduces body weight. In preclinical models and early clinical studies, the combination of ARO-ALK7 and tirzepatide has demonstrated additive effects in reducing body weight and fat mass, with ARO-ALK7 counteracting lean mass loss observed with tirzepatide alone[2][4][5][6].
