This combination therapy consists of an aromatase inhibitor (specifically anastrozole or letrozole) and recombinant human growth hormone (somatropin). It is primarily investigated for the treatment of idiopathic short stature (ISS) in adolescent boys. The therapeutic strategy employs a dual mechanism: growth hormone directly promotes linear bone growth by activating the growth hormone receptor and stimulating IGF-1 production, while the aromatase inhibitor blocks the conversion of androgens into estrogens. By lowering systemic estrogen levels, the therapy delays the fusion of the epiphyseal growth plates, thereby extending the duration of the pubertal growth spurt and increasing the predicted adult height. This specific regimen was the subject of a Phase 3 clinical trial conducted by Nemours Children's Health System.
