Overview

**ARRY-520 + bortezomib** is a combination of two investigational agents tested primarily in relapsed or refractory multiple myeloma. ARRY-520 (also called filanesib) is a highly selective, first-in-class **kinesin spindle protein (KSP)** inhibitor, inducing mitotic arrest and apoptosis in cancer cells. In preclinical studies, combining filanesib with **bortezomib**, a **proteasome inhibitor**, resulted in synergistic apoptotic activity, even in bortezomib-resistant cell lines. Early-phase clinical trials have explored this combination to improve efficacy in patients with advanced multiple myeloma, based on mechanistic rationale and evidence of activity in heavily pretreated or refractory myeloma populations[1][3].
