Arsenic trioxide + all-trans retinoic acid + ascorbic acid (AAA) is a chemotherapy-free oral regimen for the treatment of acute promyelocytic leukemia (APL). This combination targets the PML-RARA fusion protein characteristic of APL. Arsenic trioxide and all-trans retinoic acid work cooperatively to block the action of proteins needed by leukemia cells to survive and grow, while ascorbic acid appears to enhance the efficacy of this combination. The oral formulation provides a more convenient and economical approach compared to intravenous administration, allowing for outpatient treatment with reduced hospital visits.
