ART4215 + niraparib is an investigational oral combination therapy comprising ART4215, a first-in-class small molecule inhibitor of polymerase theta (Polθ), and niraparib, a poly (ADP-ribose) polymerase (PARP) inhibitor. ART4215 targets cancers with defects in DNA repair, while niraparib inhibits the activity of PARP enzymes, thereby hindering single-strand DNA repair. This combination aims to exploit synthetic lethality in cancers, particularly those with BRCA mutations or other DNA repair pathway deficiencies, by simultaneously inhibiting two key DNA repair mechanisms. The combination is under clinical evaluation, including dose-escalation studies and efficacy assessments in patients with advanced or metastatic solid tumors, notably BRCA-mutant breast cancer[1][3].
