Overview

ART4215 + talazoparib is an investigational combination therapy comprising two small molecule inhibitors for the treatment of BRCA-deficient breast cancer and other advanced or metastatic solid tumors. ART4215 is a first-in-class, highly selective, oral inhibitor of DNA polymerase theta (Polθ), a key enzyme involved in microhomology-mediated end joining (MMEJ), a DNA repair pathway that is upregulated in many cancers but minimally expressed in healthy tissue. Talazoparib (marketed as Talzenna) is an approved oral poly(ADP-ribose) polymerase (PARP) inhibitor that impairs single-strand DNA break repair. The rationale for combining these agents lies in their complementary mechanisms targeting distinct components of the DNA damage response; this dual inhibition aims to enhance synthetic lethality and overcome resistance to PARP inhibitors alone. The combination has shown tolerability and potential clinical benefit in early-phase studies, with ongoing phase 2 trials evaluating efficacy primarily in BRCA-deficient breast cancer[1][2][4][6].
