Artemether-lumefantrine + amodiaquine + primaquine is an investigational triple artemisinin-based combination therapy (TACT) being developed for the treatment of uncomplicated *Plasmodium falciparum* malaria. This combination aims to combat multidrug-resistant malaria by combining a standard fixed-dose artemisinin-based combination therapy (ACT), artemether-lumefantrine, with a third long-acting partner drug, amodiaquine, and the gametocytocidal agent primaquine. Artemether and amodiaquine act on the asexual blood stages of the parasite by inhibiting heme detoxification and generating cytotoxic free radicals, while primaquine targets gametocytes to reduce malaria transmission. The combination is being studied by the University of Oxford in collaboration with Mahidol University to evaluate pharmacokinetic interactions and safety.
