Overview

This is a hypothetical combination of six antimalarial agents—artemether, lumefantrine, dihydroartemisinin, piperaquine, artesunate, and mefloquine. All six are established components of World Health Organization (WHO)-recommended artemisinin-based combination therapies (ACTs) for the treatment of malaria. Each pair among these drugs forms a distinct ACT regimen with proven efficacy against Plasmodium falciparum and sometimes Plasmodium vivax malaria. These combinations act synergistically by rapidly reducing parasite biomass (via the artemisinin derivatives) and providing longer-term suppression to prevent recrudescence (via the partner drugs). However, there is no evidence or clinical precedent for combining all six agents into a single therapy; such a regimen would be highly unconventional due to overlapping mechanisms, increased risk of toxicity, drug-drug interactions, and lack of supporting safety or efficacy data[2][4][6]. The individual drugs work primarily by generating reactive oxygen species that damage parasitic proteins or by interfering with heme detoxification in the parasite.
