Overview
ONO-2506PO + riluzole is a combination therapy investigated for the treatment of amyotrophic lateral sclerosis (ALS). ONO-2506PO (arundic acid) is a small molecule astrocyte-modulating agent developed by Ono Pharmaceutical that inhibits the synthesis of S100B, a calcium-binding protein associated with neurodegeneration and neuroinflammation. Riluzole is a benzothiazole derivative and the standard of care for ALS, acting primarily by inhibiting glutamate release and blocking voltage-gated sodium channels. The combination was evaluated in Phase II clinical trials, but ONO-2506PO failed to demonstrate efficacy over placebo in patients with longer disease duration, leading to the discontinuation of its development for ALS. Arundic acid was also explored for other neurodegenerative conditions, including stroke and Alzheimer's disease, but development in those areas has also ceased.
