Overview

ASA404 + taxane is a combination of ASA404 (also known as 5,6-dimethylxanthenone-4-acetic acid or DMXAA or vadimezan), a small-molecule tumor-vascular disrupting agent (tumor-VDA), and a taxane-class chemotherapeutic agent (such as docetaxel or paclitaxel). ASA404 disrupts existing tumor vasculature leading to tumor necrosis, while taxanes inhibit cell division by stabilizing microtubules. This combination has been investigated in clinical trials for advanced cancers, including non-small cell lung cancer (NSCLC) and hormone-refractory prostate cancer, aiming to enhance antitumor efficacy through complementary mechanisms. ASA404 and taxanes have non-overlapping toxicity profiles and no significant pharmacokinetic interactions, supporting their combined clinical evaluation[1][3][2].
