ASC47 + ASC31 is a combination therapy being developed by Ascletis Pharma for the treatment of obesity. It consists of two distinct components: ASC47, an adipose-targeted, once-monthly subcutaneously injected thyroid hormone receptor beta (THRβ)-selective small molecule agonist, and ASC31, a dual glucagon-like peptide-1 receptor (GLP-1R) and glucose-dependent insulinotropic polypeptide receptor (GIPR) peptide agonist. ASC31 is part of Ascletis' Ultra-Long-Acting Platform (ULAP) for subcutaneously injectable peptides. In preclinical studies using diet-induced obese (DIO) mouse models, the combination demonstrated significant body weight reduction (44.8% over 14 days), outperforming the combination of ASC47 with tirzepatide. The therapy is designed to promote weight loss and body fat reduction while preserving muscle mass.
