ASC47 + tirzepatide

ASC47 + tirzepatide is a combination therapy being developed by Ascletis Pharma for the treatment of obesity. ASC47 is a first-in-class, adipose-targeted, once-monthly subcutaneously injected small molecule agonist selective for the thyroid hormone receptor beta (THRβ), designed to preserve muscle mass during weight loss. Tirzepatide is a dual agonist of the glucagon-like peptide-1 receptor (GLP-1R) and the glucose-dependent insulinotropic polypeptide receptor (GIPR). Preclinical data suggests the combination significantly enhances weight loss compared to tirzepatide monotherapy while maintaining a healthy body composition by reducing fat and preserving muscle. Phase I clinical studies are currently ongoing in Australia to evaluate the safety and efficacy of this combination.
