Drug intelligence / Profile preview
Scemblix + blinatumomab is an investigational chemotherapy-free combination regimen pairing the approved small-molecule kinase inhibitor asciminib (Scemblix) with the bispecific T-cell engager (BiTE) antibody construct blinatumomab (Blincyto). Asciminib is a first-in-class STAMP (Specifically Targeting the ABL Myristoyl Pocket) inhibitor that binds allosterically to the myristoyl pocket of ABL1, inducing an inactive conformation of the BCR-ABL1 fusion protein. Blinatumomab is a bispecific monoclonal antibody designed to direct T-cell-mediated cytotoxicity against CD19-positive B-lineage cells by simultaneously binding CD19 on B cells and CD3ε on T cells. This combination is being evaluated in a Phase I/II clinical trial (NCT06308588) for the treatment of patients with Philadelphia chromosome-positive acute lymphoblastic leukemia (Ph+ ALL) and lymphoid blast phase chronic myeloid leukemia (CML).
