Overview

This combination regimen is being evaluated by the SWOG Cancer Research Network for the maintenance phase of treatment in patients with newly diagnosed Philadelphia chromosome-positive (Ph+) acute lymphoblastic leukemia (ALL). The regimen consists of **asciminib**, a first-in-class STAMP (Specifically Targets the ABL Myristoyl Pocket) inhibitor; **dasatinib**, a second-generation BCR-ABL1 and SRC family tyrosine kinase inhibitor; **prednisone**, a synthetic glucocorticoid; and **methotrexate**, an antimetabolite. By combining two BCR-ABL1 inhibitors that target different sites of the kinase (the ATP-binding site and the myristoyl pocket), the regimen aims to achieve deeper molecular responses and overcome potential resistance mechanisms in Ph+ ALL, while utilizing standard maintenance chemotherapy components.
