Overview

Ascorbic acid (vitamin C) + irinotecan is a combination therapy being investigated for the treatment of colorectal cancer and other cancers. This combination leverages the potential synergistic effects between high-dose vitamin C and the chemotherapeutic agent irinotecan. High-dose vitamin C administration has been reported to exhibit antitumor effects in various cancer models, while irinotecan is a key drug in colorectal cancer treatment that works by inhibiting DNA replication during cell division. The combination has shown promising results in preclinical studies, demonstrating enhanced antitumor effects compared to either agent alone, particularly in drug-resistant cancer cell lines. The mechanism appears to involve reduction of reactive oxygen species (ROS) and inflammatory markers, increased collagen type I and caspase-1 expression, and enhanced cell death in tumor tissues.
