A combination therapy consisting of high-dose ascorbic acid (vitamin C) and temozolomide, primarily investigated for the treatment of glioblastoma and other high-grade gliomas. High-dose ascorbic acid appears to induce cytotoxicity and oxidative stress selectively in malignant cells, potentially enhancing the DNA-damaging effects of temozolomide through mechanisms mediated by redox-active iron. This combination is typically administered with ascorbic acid given intravenously at high doses while temozolomide is administered orally, often alongside radiation therapy.
