**AsiDNA + olaparib** is a drug combination under clinical investigation for the treatment of advanced solid tumors, particularly recurrent epithelial ovarian cancer, breast cancer, and metastatic castration-resistant prostate cancer in patients who have already progressed on prior PARP inhibitor therapy[1][3]. AsiDNA is a first-in-class DNA Damage Response (DDR) inhibitor that mimics double-stranded DNA breaks and disrupts repair mechanisms by acting as an agonist/decoy, activating but then diverting and depleting repair enzymes. Olaparib is a small molecule inhibitor of Poly (ADP-ribose) polymerase (PARP), impairing base excision repair and alternative non-homologous end joining repair pathways. Preclinical studies demonstrate that combining AsiDNA with olaparib produces synergistic anti-tumor activity, can more than double the complete response rate observed with PARP inhibition alone in preclinical PARP inhibitor-resistant tumor models, and prevents or reverses acquired resistance to PARP inhibitors, without increasing toxicity[3][4][5].
