ASKG315 + pembrolizumab is a combination regimen under clinical investigation for the treatment of advanced solid tumors. ASKG315 is a novel engineered cytokine prodrug, specifically an interleukin-15 (IL-15) prodrug designed for proteolytic activation and targeted immune stimulation, developed using AskGene Pharma’s SmartKine® cytokine prodrug technology. Its mechanism extends the half-life of IL-15 and improves safety by restricting activation to the tumor microenvironment, stimulating natural killer (NK) cells and CD8+ T cells for anti-tumor activity. Pembrolizumab is a monoclonal antibody that blocks programmed cell death protein 1 (PD-1), thereby enhancing T-cell–mediated immune response against tumors. The combination aims to synergistically augment antitumor immunity through checkpoint inhibition and cytokine-mediated immune cell activation.
