ASP1948 + nivolumab is an experimental combination therapy for advanced and metastatic solid tumors. ASP1948 is a fully human IgG4 monoclonal antibody targeting **neuropilin-1 (NRP1)**, which is expressed on regulatory T cells (Tregs). By antagonizing NRP1, ASP1948 aims to block ligand interactions and suppress Treg-mediated immunosuppression within the tumor microenvironment, thereby promoting anti-tumor immune activity. **Nivolumab** is a PD-1 immune checkpoint inhibitor that blocks the PD-1 pathway, enhancing T-cell-mediated anti-tumor responses. The combination is designed to potentiate anti-tumor immunity by simultaneously inhibiting Treg function and relieving T-cell exhaustion, enhancing the efficacy of immune checkpoint blockade. The combination is currently in early clinical development with ongoing Phase 1b studies in various solid tumor types, including non-small cell lung cancer, squamous cell carcinoma of the head and neck, metastatic castration-resistant prostate cancer, ovarian, pancreatic, colorectal, and breast cancers[1][2][3][4][5][7].
