ASP8232 + ranibizumab is a combination therapy evaluated for the treatment of center-involved diabetic macular edema (CI-DME). ASP8232 is a potent and specific small molecule inhibitor of vascular adhesion protein-1 (VAP-1), an enzyme involved in inflammation and vascular permeability. Ranibizumab is a monoclonal antibody fragment that inhibits vascular endothelial growth factor A (VEGF-A), thereby reducing abnormal blood vessel growth and leakage in the retina. The combination was studied to assess whether dual inhibition of VAP-1 and VEGF-A could provide additive or synergistic benefits in reducing retinal thickness and improving visual acuity in DME patients. Clinical trials found that while both drugs were generally well-tolerated, the combination did not show superior efficacy over ranibizumab monotherapy for visual acuity improvement[1][2].
