ASP8374 + pembrolizumab is a combination immunotherapy regimen consisting of ASP8374, a fully human IgG4 monoclonal antibody that antagonizes the immune checkpoint receptor TIGIT, and pembrolizumab, a monoclonal antibody checkpoint inhibitor targeting programmed death-1 (PD-1). ASP8374 blocks TIGIT-mediated inhibition of lymphocytes to enhance anti-tumor immunity, while pembrolizumab blocks PD-1 to similarly release inhibitory immune signaling. The combination is investigated for its potential to achieve greater immune activation and anti-tumor response than either agent alone, with development focused on advanced and metastatic solid tumors. Preclinical and early clinical studies have suggested synergistic or additive effects on T cell activation and anti-tumor activity with the dual blockade[3][5][7].
