Overview

ASP9853 + paclitaxel is an investigational combination therapy that pairs ASP9853, a small molecule inhibitor of inducible nitric oxide synthase (iNOS) dimerization, with paclitaxel, a taxane-class chemotherapeutic agent. ASP9853 reduces the production of nitric oxide by inhibiting iNOS, which may counteract mechanisms that make tumors less responsive to taxanes such as paclitaxel. Preclinical studies showed that ASP9853 in combination with taxanes (including paclitaxel) produced greater tumor growth inhibition than taxane alone in various xenograft models. However, in clinical development, trials combining ASP9853 with docetaxel (a taxane closely related to paclitaxel) were terminated due to toxicity and lack of clear efficacy; the planned clinical study of ASP9853 + paclitaxel was not started, with development left incomplete at the Phase 1 stage[1][3][4][5].
