This combination therapy regimen, evaluated in the C19-ACS clinical trial (NCT04333407), consists of five small molecule drugs: aspirin, clopidogrel, rivaroxaban, atorvastatin, and omeprazole. Developed by Imperial College London, the regimen was designed to prevent cardiac complications in hospitalized COVID-19 patients with pre-existing cardiovascular risk factors. The rationale behind this multi-drug approach is that COVID-19 severity is often exacerbated by direct injury to the cardiovascular system, and early acute coronary syndrome (ACS) therapy might mitigate these risks. Atorvastatin, a key component, is an HMG-CoA reductase inhibitor that provides both cholesterol-lowering and pleiotropic vascular benefits. The trial found modest evidence of a reduction in hospital length of stay, although it did not significantly reduce mortality.
