AST-201 is an investigational, intradermally administered plasmid DNA therapeutic cancer vaccine developed by Aston Sci for ovarian cancer. The pUMVC3-based construct encodes N-terminal epitopes of human insulin-like growth factor-binding protein 2, inducing IGFBP2-specific type 1 helper T-cell, memory T-cell, and cytotoxic T-cell immune responses against IGFBP2-expressing tumor cells. Aston Sci's CornerStone-004 program is a randomized Phase 2 study in newly diagnosed homologous-recombination proficient Stage III ovarian cancer after debulking surgery, evaluating AST-201 with rhuGM-CSF and adjuvant paclitaxel plus carboplatin; the sponsor describes the Phase 2 IND as approved and the study as site-ready but not opened.
