Overview

AT-527 is a novel modified guanosine nucleotide prodrug inhibitor of the hepatitis C virus (HCV) NS5B polymerase. It is designed to have increased in vitro antiviral activity and improved metabolic stability compared to earlier guanosine analogs. AT-527 does not require activation by cytochrome P450 enzymes and demonstrates enhanced potency against all HCV genotypes, especially genotype 3[6]. Carbamazepine is an anticonvulsant medication used primarily for the treatment of epilepsy (various seizure types), trigeminal neuralgia (nerve pain), and bipolar disorder. Its mechanism involves inhibition of voltage-dependent sodium channels in neuronal membranes, stabilizing hyperexcited nerve cells and reducing synaptic transmission[4][5][8]. The combination "at-527 + carbamazepine" refers to co-administration or study of both drugs together; there are clinical studies investigating their pharmacokinetic interactions[3].
