Overview

AT-7687 + amylin is a combination therapy under development by Antag Therapeutics for the treatment of obesity. The combination consists of AT-7687, a first-in-class peptide-based gastric inhibitory polypeptide receptor (GIPR) antagonist, and an amylin analog (specifically cagrilintide, a dual amylin and calcitonin receptor agonist). AT-7687 is designed to block GIPR signaling, which is genetically and clinically validated to reduce fat storage, improve insulin sensitivity, and promote weight loss without the gastrointestinal side effects typical of GLP-1 receptor agonists. When combined with an amylin analog, the therapy leverages synergistic metabolic pathways to enhance weight loss, improve body composition (preferential fat loss over lean mass), and significantly boost insulin sensitivity. The combination has completed preclinical proof-of-concept studies in non-human primates, demonstrating robust and sustained weight loss.
