Drug intelligence / Profile preview
The combination of atebimetinib, an anti-PD-1 antibody, carboplatin, and pemetrexed is an investigational therapeutic regimen being developed for the first-line treatment of patients with advanced or metastatic RAS-mutant non-small cell lung cancer (NSCLC). Atebimetinib (IMM-1-104) is an oral, once-daily Deep Cyclic Inhibitor of MEK designed to provide pulsatile, deep inhibition of the MAPK pathway, which prevents tumor resistance and minimizes toxicities while reversing MAPK-driven immune suppression. The anti-PD-1 component (specifically cemiplimab, provided via a clinical supply agreement with Regeneron) acts as an immune checkpoint inhibitor to restore T-cell-mediated anti-tumor immunity. Carboplatin and pemetrexed serve as a standard platinum-doublet chemotherapy backbone, inducing cytotoxic tumor cell death by damaging DNA and inhibiting folate-dependent nucleotide synthesis, respectively. Preclinical studies in lung cancer models have demonstrated that adding atebimetinib to anti-PD-1 and chemotherapy significantly improves median survival compared to anti-PD-1 plus chemotherapy alone.
