Overview

Atezolizumab + azacitidine is a pharmaceutical combination consisting of atezolizumab, a humanized monoclonal antibody that targets programmed death-ligand 1 (PD-L1), and azacitidine, a hypomethylating agent and cytidine analogue. Atezolizumab blocks PD-L1 on tumor and immune cells, restoring T-cell mediated antitumor immunity by preventing inhibition of activated T cells and promoting immune response[2][4][6]. Azacitidine incorporates into DNA and RNA, inhibiting DNA methyltransferase, leading to DNA hypomethylation and direct cytotoxicity to abnormal hematopoietic cells[8]. This combination has been tested primarily for myelodysplastic syndromes (MDS), specifically in HMA-naïve and relapsed/refractory patients, aiming to enhance antitumor immune effects and epigenetic modulation[1][3][9].
