Overview

This is a four-drug combination regimen consisting of atezolizumab, bevacizumab, carboplatin, and pemetrexed. - **Atezolizumab** is a monoclonal antibody immunotherapy that targets programmed death-ligand 1 (PD-L1), blocking its interaction with PD-1 and B7.1 receptors on T cells and antigen-presenting cells. This inhibition enhances the immune system's ability to detect and destroy cancer cells. - **Bevacizumab** is a monoclonal antibody that inhibits vascular endothelial growth factor A (VEGF-A), preventing angiogenesis (the formation of new blood vessels) necessary for tumor growth. - **Carboplatin** is a platinum-based chemotherapy agent that causes DNA crosslinking and damage, leading to apoptosis in rapidly dividing cancer cells. - **Pemetrexed** is an antifolate chemotherapy agent that disrupts folate-dependent metabolic processes essential for cell replication. This combination has been studied primarily in advanced non-small cell lung cancer (NSCLC) as first-line therapy for patients with non-squamous histology who are not candidates for curative surgery or radiation. The regimen leverages both immunotherapeutic mechanisms (atezolizumab) and antiangiogenic effects (bevacizumab), along with cytotoxic chemotherapy agents targeting DNA synthesis and repair pathways.
