Overview

This is a combination therapy regimen consisting of two monoclonal antibodies (atezolizumab and bevacizumab) and two chemotherapy agents (gemcitabine and oxaliplatin). Atezolizumab is an immune checkpoint inhibitor targeting programmed death-ligand 1 (PD-L1), while bevacizumab is an anti-angiogenic agent targeting vascular endothelial growth factor (VEGF). Gemcitabine and oxaliplatin (GEMOX) are standard cytotoxic chemotherapies. The combination is being investigated for the treatment of advanced biliary tract cancer (BTC), including intrahepatic cholangiocarcinoma, extrahepatic cholangiocarcinoma, and gallbladder cancer. The rationale is that anti-VEGF therapy can normalize tumor vasculature and enhance the infiltration of immune cells, potentially synergizing with PD-L1 inhibition and chemotherapy-induced immunogenic cell death.
