Overview

This is a combination regimen of three drugs: atezolizumab (a monoclonal antibody targeting PD-L1), bevacizumab (a monoclonal antibody targeting VEGF-A), and pemetrexed (a folate antimetabolite). Atezolizumab acts as an immune checkpoint inhibitor by blocking the interaction between PD-L1 and its receptors, thereby enhancing T-cell mediated anti-tumor response. Bevacizumab inhibits angiogenesis by binding to vascular endothelial growth factor A (VEGF-A), preventing it from activating its receptors on endothelial cells. Pemetrexed is a multi-targeted antifolate that disrupts folate-dependent metabolic processes essential for cell replication, inhibiting enzymes involved in purine and pyrimidine synthesis. This combination is primarily used in the treatment of metastatic non-squamous non-small cell lung cancer (NSCLC) without EGFR or ALK genomic aberrations[3][4][7].
