Overview

Atezolizumab + bevacizumab + tocilizumab is an experimental combination therapy comprising three monoclonal antibodies: atezolizumab, an anti-PD-L1 checkpoint inhibitor; bevacizumab, a VEGF inhibitor; and tocilizumab, an IL-6 receptor antagonist. Atezolizumab binds to and inhibits PD-L1 on tumor and immune cells, enhancing antitumor immune responses. Bevacizumab binds vascular endothelial growth factor (VEGF), inhibiting angiogenesis necessary for tumor growth. Tocilizumab targets the interleukin-6 receptor (IL-6R), blocking IL-6-mediated signaling associated with inflammation and cancer progression. This triplet strategy aims to augment antitumor immune responses while modulating the tumor microenvironment and addressing resistance pathways, especially where immunotherapy or anti-angiogenic monotherapy are insufficient. Clinical trials have investigated this combination in solid tumors, including studies of stereotactic radiation with this regimen in advanced cancers[6].
