Atezolizumab + cobimetinib is an investigational combination therapy consisting of atezolizumab, a monoclonal antibody targeting programmed death-ligand 1 (PD-L1), and cobimetinib, a small molecule inhibitor of mitogen-activated protein kinase kinase (MEK). Atezolizumab acts as an immune checkpoint inhibitor by blocking PD-L1 and enhancing T-cell–mediated antitumor responses. Cobimetinib inhibits MEK1/2 in the MAPK pathway, suppressing tumor cell proliferation and survival. The combination aims to enhance tumor immunogenicity while directly inhibiting oncogenic signaling pathways. This regimen has been studied in advanced solid tumors including biliary tract cancer and melanoma[2][3][4][5].
