Atezolizumab + cyclophosphamide + vinorelbine is an investigational combination regimen comprising three agents with distinct mechanisms of action, used primarily in oncology clinical trials. Atezolizumab is a monoclonal antibody that targets programmed death-ligand 1 (PD-L1), blocking its interaction with PD-1 and B7.1 receptors to enhance anti-tumor immune responses. Cyclophosphamide is an alkylating agent that crosslinks DNA, leading to cell death, and can also modulate the immune system at low doses. Vinorelbine is a semi-synthetic vinca alkaloid that inhibits microtubule assembly by binding tubulin, resulting in mitotic arrest and apoptosis of rapidly dividing cells. This combination has been explored for synergistic effects in various cancers, including non-small cell lung cancer (NSCLC) and sarcomas[1][2][6]. The regimen aims to combine direct cytotoxicity from chemotherapy with immunomodulation from atezolizumab.
