Overview

Atezolizumab + daratumumab + pomalidomide is an experimental multi-drug combination therapy consisting of three distinct agents: atezolizumab, daratumumab, and pomalidomide. - **Atezolizumab** is a monoclonal antibody targeting programmed cell death-ligand 1 (PD-L1) to inhibit immune checkpoint signaling, thereby promoting anti-tumor T-cell responses. - **Daratumumab** is a monoclonal antibody targeting CD38, a glycoprotein highly expressed on multiple myeloma cells, inducing cell death via multiple immune-mediated mechanisms. - **Pomalidomide** is a third-generation immunomodulatory agent that works via cereblon-mediated modulation of E3 ubiquitin ligase activity to enhance T-cell and natural killer cell activation and suppress pro-inflammatory cytokines. This combination is investigational for the treatment of relapsed/refractory multiple myeloma and/or other hematologic malignancies. There is no evidence that this particular three-drug combination is FDA approved. Most clinical trial data and approvals involve the two-drug combination daratumumab + pomalidomide (often with dexamethasone), while some studies have investigated experimental combinations potentially including immune checkpoint inhibitors[2][3][4][5].
