Overview

**Atezolizumab + durvalumab** is a combination of two monoclonal antibody drugs, each targeting programmed cell death 1 ligand 1 (PD-L1), used in oncology as immune checkpoint inhibitors. Both atezolizumab (Tecentriq) and durvalumab (Imfinzi) bind to PD-L1, preventing its interaction with PD-1 and CD80 on T cells, thereby inhibiting immune checkpoint signaling. This blockade releases T cell suppression, enhances cytotoxic T cell activation, and restores immune-mediated antitumor activity. Both drugs are typically used intravenously for the treatment of lung cancer and other solid tumors, particularly extensive-stage small cell lung cancer (ES-SCLC), where they have shown survival benefits when added to standard platinum-etoposide chemotherapy. Atezolizumab is developed by Genentech (Roche) and durvalumab by AstraZeneca. As a dual PD-L1 blockade combination, this regimen aims to maximize anti-tumor immunity.
