Atezolizumab + enzalutamide is a combination regimen consisting of **atezolizumab**, a humanized monoclonal antibody inhibitor of programmed death-ligand 1 (PD-L1), and **enzalutamide**, an oral androgen receptor inhibitor. This combination was investigated primarily in metastatic castration-resistant prostate cancer (mCRPC), based on the hypothesis that enzalutamide might enhance interferon-gamma signaling and thus sensitize tumor cells to immune-mediated cytotoxicity induced by PD-L1/PD-1 inhibition. The combination was studied in the phase 3 IMbassador250 trial in patients with mCRPC after failure of androgen synthesis inhibitors and a taxane regimen. The trial found no added efficacy with this combination versus enzalutamide alone in unselected patients.
