Drug intelligence / Profile preview
Tecentriq + fluorouracil + leucovorin + oxaliplatin is a chemo-immunotherapy regimen combining the programmed death-ligand 1 (PD-L1) blocking monoclonal antibody atezolizumab (Tecentriq) with the modified FOLFOX6 (mFOLFOX6) chemotherapy regimen (consisting of the fluoropyrimidine antimetabolite fluorouracil, the folate analog leucovorin, and the platinum-based DNA-damaging agent oxaliplatin). Developed by Genentech (a member of the Roche Group), this combination is indicated for the adjuvant treatment of adult and pediatric patients 2 years of age and older with Stage III mismatch repair-deficient (dMMR) colon cancer. The regimen works synergistically: the chemotherapy components induce immunogenic cell death and tumor debulking, while atezolizumab blocks the PD-1/PD-L1 pathway to reactivate T-cell-mediated anti-tumor immune responses. In the Phase III ATOMIC trial, the addition of atezolizumab to adjuvant mFOLFOX6 significantly improved disease-free survival compared to chemotherapy alone, reducing the risk of disease recurrence or death by 50%.
