Atezolizumab + fulvestrant + ipatasertib is an investigational combination therapy comprising three agents with distinct mechanisms of action, evaluated primarily in breast cancer. Atezolizumab is a monoclonal antibody immune checkpoint inhibitor targeting programmed death-ligand 1 (PD-L1), enhancing anti-tumor immune responses. Fulvestrant is a selective estrogen receptor degrader that binds to and accelerates the degradation of the estrogen receptor, blocking estrogen signaling in hormone receptor–positive cancers. Ipatasertib is a small molecule inhibitor of AKT, a key kinase in the PI3K/AKT pathway involved in cell survival and proliferation. This triplet regimen aims to target hormone-driven growth, oncogenic signaling via AKT, and tumor immune evasion simultaneously.
