Overview

Atezolizumab + ipatasertib + paclitaxel is a triplet combination therapy being investigated primarily for the treatment of triple-negative breast cancer (TNBC). This combination brings together three distinct mechanisms of action: atezolizumab is a PD-L1 inhibitor (immune checkpoint inhibitor), ipatasertib is an AKT inhibitor, and paclitaxel is a taxane chemotherapy agent that stabilizes microtubules. The triplet has been studied in clinical trials including the phase III IPATunity170 trial for locally advanced or metastatic TNBC. The regimen typically consists of intravenous atezolizumab 840 mg (days 1 and 15), oral ipatasertib 400 mg/day (days 1-21), and intravenous paclitaxel administered in a 28-day cycle.
