Atezolizumab + ramucirumab is a combination therapy that pairs two distinct mechanisms of action to treat non-small cell lung cancer (NSCLC). Atezolizumab is a humanized IgG antibody that binds to PD-L1, preventing its interaction with PD-1 and B7-1, thereby enhancing T-cell immune response against cancer cells. Ramucirumab is a human monoclonal antibody that targets VEGFR-2, inhibiting angiogenesis by blocking the binding of VEGF ligands (VEGF-A, VEGF-C, and VEGF-D). This combination aims to simultaneously activate the immune system against cancer cells while disrupting tumor blood supply. Clinical studies have shown that while the overall response rate may be modest, the combination demonstrates a high clinical benefit rate and promising overall survival in heavily pretreated NSCLC patients who have progressed after platinum-based chemotherapy and immune checkpoint inhibitors.
