Overview

Atezolizumab + talazoparib is an investigational combination therapy being studied primarily for the treatment of metastatic triple-negative breast cancer (mTNBC), particularly in patients with PD-L1 positive tumors. Atezolizumab is a monoclonal antibody that acts as an immune checkpoint inhibitor by targeting programmed death-ligand 1 (PD-L1), thereby enabling T-cell activation and enhancing anti-tumor immunity. Talazoparib is a potent small molecule inhibitor of poly (ADP-ribose) polymerase (PARP) enzymes, specifically PARP1 and PARP2, which are involved in DNA repair. By inhibiting these enzymes, talazoparib prevents the repair of single-strand DNA breaks, leading to synthetic lethality in tumor cells with deficient homologous recombination repair mechanisms. Preclinical studies suggest that combining these agents may enhance antitumor activity regardless of BRCA mutation status[1][4][5][6].
