This is a multi-agent investigational combination therapy comprising: - **Atezolizumab**: a monoclonal antibody that inhibits programmed death-ligand 1 (PD-L1), thereby restoring and amplifying T-cell–mediated antitumor immune responses. - **Tiragolumab**: a monoclonal antibody targeting TIGIT, an inhibitory receptor on T cells and natural killer cells, designed to further enhance antitumor immunity; often trialed in combination with atezolizumab. - **Cisplatin**: a platinum-based small molecule chemotherapy agent that causes crosslinking of DNA, leading to apoptosis in cancer cells. - **5-Fluorouracil (5FU)**: a pyrimidine analog functioning as an antimetabolite, inhibiting thymidylate synthase and interfering with DNA synthesis. This combination aims to leverage both immune checkpoint inhibition and cytotoxic chemotherapy for synergistic antitumor efficacy, particularly in advanced solid tumors such as non-small cell lung cancer, esophageal cancer, and head and neck cancers[1][2][4][5][6][7][8]. The regimen is currently under investigation in phase I-III trials for indications including metastatic non-small cell lung cancer and esophageal carcinoma, primarily in immunotherapy-naive or previously treated patient populations.
