Overview

ATG-017 + nivolumab is a combination therapy under investigation for advanced solid tumors. ATG-017 is an orally administered, potent, and selective small molecule inhibitor of extracellular signal-regulated protein kinases 1 and 2 (ERK1/2), developed by Antengene. Nivolumab is a human monoclonal antibody that targets the programmed cell death protein 1 receptor (PD-1) on activated T-cells, functioning as an immune checkpoint inhibitor; it is developed and marketed by Bristol Myers Squibb under the brand name Opdivo. The combination aims to leverage synergistic effects observed in preclinical models, particularly improving efficacy in tumors resistant to immune checkpoint inhibitors. The ongoing ERASER study evaluates safety, tolerability, pharmacokinetics, and preliminary efficacy of this combination in patients with advanced solid tumors[1][3][4][8].
