ATN-224 + bortezomib is a combination of two investigational and approved drugs studied primarily for the treatment of multiple myeloma. ATN-224, also known as choline tetrathiomolybdate, is a copper chelator that inhibits superoxide dismutase 1 (SOD1) activity, increases intracellular oxidants, and induces peroxynitrite-dependent cell death by modulating the cellular redox environment and causing mitochondrial dysfunction. It targets SOD1 and cytochrome c oxidase in mitochondria[2]. Bortezomib is an FDA-approved proteasome inhibitor used to treat multiple myeloma and mantle cell lymphoma. It works by blocking proteasomes—enzymes that break down proteins inside cells—leading to accumulation of proteins in cancer cells and subsequent cell death[1][3]. The combination has been investigated in clinical trials for its potential synergistic effects against hematological malignancies such as multiple myeloma[6].
